Transients are common in smooth muscle, yet are difficult to measure accurately because of the problems caused by their stochastic occurrence, over an often wide field of view, in an organ that it prone to contract. To overcome this problem, we've developed a series of imaging protocols and analysis routines to acquire and then analyse, in an automated fashion, the frequency, location and amplitude of such events. While this approach may be applied in other contexts, our own work involves the detection of local purinergic Ca transients allow the optical monitoring of neurotransmitter release in a manner that has many advantages over electrophysiology. Apart from the greatly improved spatial resolution, optical recording has the additional advantage of allowing the recording of transmitter release from many distinguishable sites simultaneously. Furthermore, the optical plane of focus is easier to maintain or correct during long recording series than is the repositioning of an intracellular sharp microelectrode.

Personally, I consider most of the new cameras released in the past few years as almost being niche cameras, that is, their capabilities are best utilized by photographers shooting specific genres or needing specific capabilities or... Great photographs are most often the products of incredible photographers. First, let me state that without having tried this camera out -- and I likely never will as I'm a Canon guy -- the Nikon D4 is probably an incredible piece of technology. If you're a sports photographer, for instance, cameras with big buffers capable of capturing images as fast or faster than a machine gun spits bullets might be perfect for you. Let me follow that statement by reminding everyone that great photos are not generally the products of incredible technologies....

Germany's Max Planck Society and the microscope maker Leica Microsystems have joined forces to commercialize a new technique to allow optical microscopes to look at even smaller objects than was previously possible. The sample is viewed through a confocal microscope, which uses a tiny aperture to ensure that light is only gathered from a very small part of the sample. This latest method is based on stimulated emission depletion (STED) microscopy, which is a technique for beating the diffraction limit that was first demonstrated more than a decade ago by Max Planck researcher Stefan Hell. However, it does have one major limitation: it cannot normally resolve objects smaller than about 200 nm, the so-called diffraction limit. A major breakthrough came about a decade ago when Hell and colleagues at the Max Planck Institute for Biophysical Chemistry in Göttingen, Germany, invented STED. The method involves fluorescence microscopy, whereby a molecule of interest is tagged with a dye that gives off light at a specific colour when illuminated with a spot of light.
